I have got a letter for asking my availability for skype interview, how could i reply to them that i am interested and available on a specific time.

Replying for an email is not a hassle anymore, first, the starter of the email has indicated all the necessary parts of the email, like addresses, and names, all you need to do is to provide your position and name at the end with your own personal reply just like chatting with another man but maintain the formality, Emails now are fast enough that it doesn`t need a formal format for replying.
However, in some higher cases, people are using signatures to introduce themselves at the end of every response on emails, you can also try that.
You can reply in their email invitation with simple words but in the exact wordings. That If you wanted to be in schedule interview, then why not. Just write like this.

Sir/Ma`am as much as I wanted to appear in the interview it is unfortunate though because I would be unable to appear on the interview date you have set. (discuss your reasons why, make it sure though that your reason is valid and justifiable).

First of all, thank the sender for the opportunity at the interview. You should list the days and hours that you are available and schedule something that is convenient for both of you.
Write down your skype ID and ask for theirs also so you can add to your contact list.
I would call the company that set the interview up with you by email on the phone. It`s more personable. They will accommodate your request, and you don`t have to worry about creating an email where you might make a typo or something.
